Usually, you will have two knobs on the base of your microscope. One is the coarse focus knob and the other is the fine focus knob. These knobs will help you focus and adjust the specimen under different magnifications. The coarse focus knob will help you focus on the specimen at low power, while the fine focus knob will help you focus on the specimen under higher power.
The coarse focus knob should never be used to focus closer to the object while looking through the eyepiece. If you try this, you will be focusing on the object behind your microscope and it will move further out of focus. Also, you can damage the focusing knob if you try to turn it too far. To focus, you should turn the knob a few millimeters at a time and the specimen will be in focus.
The coarse focus knob moves the stage a larger distance per rotation than the fine focus knob. This helps to get the image in focus faster. You can also use the coarse knob to lower the stage. When you are looking at the specimen through the eyepiece, you will need to adjust the light intensity. There is a light intensity knob on the base of your microscope. You will also need to adjust the light intensity at each magnification you use. For example, you will need to adjust the light intensity when you use a 10X objective.
